GENETIC DISSECTION OF lin-11 REGULATION IN DIFFERENTIATION OF C. elegans AMPHID SENSORY NEURONS
TITLE: Genetic dissection of lin-11 regulation in differentiation of C. elegans amphid sensory neurons ! ! iii! This thesis is dedicated to my family. Abstract The expression of lin-11 is regulated by enhancers located upstream from, as well as within, lin-11 intronic sequences. Multiple regulatory inputs control the spatiotemporal expression pattern of lin-11. To better understand that process...
متن کاملTranscriptional control of cell-cycle quiescence during C. elegans development.
During the development of the C. elegans reproductive system, cells that give rise to the vulva, the vulval precursor cells (VPCs), remain quiescent for two larval stages before resuming cell division in the third larval stage. We have identified several transcriptional regulators that contribute to this temporary cell-cycle arrest. Mutation of lin-1 or lin-31, two downstream targets of the Rec...
متن کاملIntronic Alternative Splicing Regulators Identified by Comparative Genomics in Nematodes
Many alternative splicing events are regulated by pentameric and hexameric intronic sequences that serve as binding sites for splicing regulatory factors. We hypothesized that intronic elements that regulate alternative splicing are under selective pressure for evolutionary conservation. Using a Wobble Aware Bulk Aligner genomic alignment of Caenorhabditis elegans and Caenorhabditis briggsae, w...
متن کاملExpression patterns of intronic microRNAs in Caenorhabditis elegans
BACKGROUND MicroRNAs (miRNA) are an abundant and ubiquitous class of small RNAs that play prominent roles in gene regulation. A significant fraction of miRNA genes reside in the introns of the host genes in the same orientation and are thought to be co-processed from the host gene mRNAs and thus depend on the host gene promoter for their expression. However, several lines of evidence for indepe...
متن کاملPatterns of flanking sequence conservation and a characteristic upstream motif for microRNA gene identification.
MicroRNAs are approximately 22-nucleotide (nt) RNAs processed from foldback segments of endogenous transcripts. Some are known to play important gene regulatory roles during animal and plant development by pairing to the messages of protein-coding genes to direct the post-transcriptional repression of these messages. Previously, we developed a computational method called MiRscan, which scores f...
